目的探索肝癌患者组织中miR-122、miR-375的表达情况。方法选取2013—2014年河南省省直第二医院、郑州大学第二附属医院的原发肝癌手术切除患者30例。提取各癌组织标本中的miRNA(以癌旁正常组织为对照),定量逆转录PCR的方法检测miR-122、miR-375的相对表达水平,并分析各组各组织中miR-122、miR-375表达水平及与肝癌病理学特征的关系。结果 30例肝癌组织中miR-122、miR-375相对表达水平分别为(16.84±4.11)和(14.32±3.81),均都显著高于癌旁的正常组织,差异非常有显著性(P<0.01);miR-122的相对表达水平与肝癌的大小、肿瘤的分化程度及其TNM分期显著相关(P<0.05);而miR-375相对的表达水平则与患者年龄和性别及肝癌的大小、分化程度、TNM分期均无关(P>0.05)。结论肝癌组织中miR-122、miR-375表达升高,且miR-122的相对表达水平和肝癌大小、肿瘤的分化程度及TNM分期有显著相关。
Objective To explore the expression of miR-122 and miR-375 in HCC tissue. Methods Thirty patients with primary liver cancer who underwent surgical resection from 2013 to 2014 in Henan Provincial Second Hospital and Zhengzhou Second Affiliated Hospital were selected. The relative expression levels of miR-122 and miR-375 were detected by quantitative reverse transcriptase-polymerase chain reaction (RT-PCR). The expressions of miR-122, miR- 375 and its relationship with pathological features of liver cancer. Results The relative expression levels of miR-122 and miR-375 in HCC tissues were (16.84 ± 4.11) and (14.32 ± 3.81), respectively, which were significantly higher than those in normal tissues (P <0.01) ). The relative expression level of miR-122 was significantly associated with the size of hepatocellular carcinoma, tumor differentiation and TNM stage (P <0.05), while the relative expression level of miR-375 was related to the age and sex of patients and the size and differentiation of HCC Degree and TNM stage (P> 0.05). Conclusion The expression of miR-122 and miR-375 in HCC tissue is increased, and the relative expression level of miR-122 is significantly correlated with the size of HCC, tumor differentiation and TNM stage.
