①目的检测人巨细胞病毒特异性转移因子(HCMV-STF)对小鼠脾细胞分化增殖作用。②方法采用微量细胞培养法培养小鼠脾细胞,共分5组,分别加入1640培养液(空白对照),PHA,HCMV-Ag,HCMV-STF,HCMV-Ag+HCMV-STF.培养48h后加入氚-胸腺嘧啶(3H-TDR),再培养24h,收集细胞,用液体闪烁计数器计数(cpm),计算促进率。③结果HCMV-Ag+HCMV-STF组与HCMV-Ag,HCMV-STF组比较具有明显的促进小鼠脾细胞分化增殖的作用(q=3.35,4.75,P<0.05)。④结论HCMV-STF具有相应抗原的依赖活性;同时也证明3H-TDR掺入法检测小鼠脾细胞分化增殖效应,是特异性转移因子质控的一种较为理想的测试方法。
Objective To detect the effect of human cytomegalovirus-specific transfer factor (HCMV-STF) on the proliferation and differentiation of mouse spleen cells. MethodsMice spleen cells were cultured by micro-cell culture and divided into 5 groups. The cells were cultured in 1640 medium (PHA, HCMV-Ag, HCMV-STF and HCMV-Ag + HCMV-STF respectively). Tritiated-thymidine (3H-TDR) was added after 48 h of cultivation, and cultured for a further 24 h. Cells were harvested and counted by liquid scintillation counter (cpm) to calculate the promotion rate. Results Compared with HCMV-Ag and HCMV-STF groups, HCMV-Ag + HCMV-STF group had a significant effect of promoting the proliferation and differentiation of mouse spleen cells (q = 3.35, 4.75, P <0.05). Conclusion HCMV-STF has antigen-dependent activity. At the same time, it also proved that 3H-TDR incorporation method can detect the differentiation and proliferation of mouse spleen cells, which is an ideal test method for the quality control of specific transfer factor.
