                                目的建立高效液相色谱法测定榴莲中姜黄素、脱甲氧基姜黄素和双脱甲氧基姜黄素3种姜黄素类物质的含量的分析方法。方法样品经甲醇超声提取,以0.4%乙酸水溶液-乙腈为流动相进行等度洗脱,经TSKgel ODS柱进行分离,采用二极管阵列检测器在425 nm处检测,以外标法定量。结果姜黄素类化合物在2.5~200 ng/m L的浓度范围内线性关系良好,相关系数r>0.99。姜黄素类化合物的检测限均为10μg/kg,在低、中、高3个加标水平下,榴莲肉中化合物回收率均大于70%,相对标准偏差均低于10%(n=6)。结论该方法快速准确,可适用于榴莲中姜黄素类化合物的检测。
Objective To establish an HPLC method for the determination of curcumin, demethoxy curcumin and didemethoxy curcumin in curcumin. Methods The sample was extracted by methanol and eluted with 0.4% acetic acid in acetonitrile as mobile phase. The sample was separated by TSKgel ODS column and detected by using a diode array detector at 425 nm and quantified by external standard method. Results Curcumin showed a good linearity in the concentration range of 2.5 ~ 200 ng / m L with a correlation coefficient r> 0.99. The detection limits of curcumin were all 10μg / kg. The recoveries of the compounds in durian meat were all above 70% with the relative standard deviations below 10% (n = 6) at the three spiked levels of low, medium and high, . Conclusion The method is rapid and accurate and can be applied to the detection of curcuminoids in durian.
