为提高大白菜小孢子胚成苗率,对影响胚发育成苗的培养基水分状况进行了研究。在诱导胚胎发生的NLN13培养基中,培养2~3周形成的子叶期胚质量好,成苗率高;随着培养时间的延长,胚质量和成苗率逐渐降低。诱导胚发育成苗的MS0培养基的琼脂含量,对于小孢子胚的成苗率有重要影响。将胚培养基琼脂含量从0.8%提高到1.2%,成苗率从37.5%提高到85.8%,同时玻璃化和二次分化再生现象均明显减少,再生植株健壮。
In order to improve the microspore embryogenesis rate of Chinese cabbage, the moisture condition of the culture medium that affected embryo development was studied. In the induced embryogenesis of NLN13 medium, the cotyledonary embryos formed during 2 ~ 3 weeks of culture had good quality and high seedling rate. With the prolongation of culture time, the embryo quality and seedling emergence rate decreased gradually. The agar content of MS0 medium inducing embryogenesis has an important effect on the seedling rate of microspore embryos. The embryo medium agar content increased from 0.8% to 1.2%, the seedling rate increased from 37.5% to 85.8%, while the vitrification and secondary differentiation of regeneration were significantly reduced, regenerated plants robust.
