采用反转录PCR和c DNA末端快速扩增技术从春兰(Cymbidium goeringii)中分离到1个B类MADS-box基因CgDEF4基因。CgDEF4的基因登录号为KU058678,属于PaleoAP3组的PeMADS4类基因,含有1个678 bp的开放阅读框(open reading frame,ORF),分子量24.05 k D,编码225个氨基酸。实时荧光定量表达分析表明,CgDEF4在春兰正常花和蝶花各花器官中差异表达。CgDEF4在正常花的唇瓣中强烈表达,在侧瓣的表达量较低;而在蝶花中在唇瓣和唇瓣化的侧瓣中都强烈表达,说明CgDEF4基因可能在春兰蝶花侧瓣唇瓣化的过程中具有重要作用。
One class B MADS-box gene CgDEF4 gene was isolated from Cymbidium goeringii by reverse transcription PCR and c DNA rapid amplification. The gene accession number of CgDEF4 is KU058678. The PeMADS4 gene belonging to the PaleoAP3 group contains a 678 bp open reading frame (ORF) with a molecular weight of 24.05 kD and encodes a polypeptide of 225 amino acids. Real-time fluorescence quantitative analysis showed that CgDEF4 was differentially expressed in floral organs of normal flowers and butterflies. CgDEF4 was strongly expressed in the normal flower lip and lower in the lateral flap, whereas CgDEF4 was strongly expressed in both the lip and the labial flank, indicating that the CgDEF4 gene may be expressed in the leaf Lip process has an important role.
