目的:应用反相高效液相色谱/二极管阵列检测(RP-HPLIC-DAD)测定中药降糖制剂中可能存在的3种化学药品。方法:采用RP-HPLC-DAD对临床常用3种降糖化学药品进行筛选,并对降糖中药制剂进行初步分析。色谱柱:Agilent ZORB- AX ODS C18柱(4.6 mm×250 mm,5μm);流动相:醋酸盐缓冲液(10 mmol·L-1醋酸胺,20mmol·L-1三乙胺,用醋酸调节pH 5.5)与乙腈,梯度洗脱,流速:0.6 ml·min-1,检测波长230 nm,格列吡嗪为内标物进行定量分析。结果:所测定的中药降糖药中含有三种化学药品组分,分别为格列本脲、苯乙双胍、阿卡波糖,并能在该色谱条件下与中药完全分离。结论:本方法可用于所测中药降糖药中3种化学药品成分的分析。
Objective: To use RP-HPLIC-DAD to determine the possible three kinds of chemicals in the traditional Chinese medicine hypoglycemic agent. Methods: Three kinds of hypoglycemic chemical drugs commonly used in clinical screening were selected by RP-HPLC-DAD, and preliminary analysis of hypoglycemic Chinese medicine preparations was conducted. Column: Agilent ZORB-AX ODS C18 column (4.6 mm×250 mm, 5 μm); mobile phase: acetate buffer (10 mmol·L-1 acetate, 20 mmol·L-1 triethylamine, used Acetic acid was adjusted to pH 5.5) with acetonitrile, gradient elution, flow rate: 0.6 ml·min-1, detection wavelength was 230 nm, and glipizide was used as an internal standard for quantitative analysis. Results: The traditional Chinese medicine hypoglycemic agents contained three chemical components, namely glibenclamide, phenformin, and acarbose, and they can be completely separated from traditional Chinese medicine under the chromatographic conditions. Conclusion: This method can be used for the analysis of the three chemical constituents of traditional Chinese medicine hypoglycemic agents.
