目的:应用激素联合内毒素脂多糖建立兔股骨头坏死(osteonecrosis of the femoral head,ONFH)模型。方法:将40只4月龄新西兰大白兔随机分成2组,每组20只,A组为对照组,B组为造模组。第1周B组开始造模,采用大剂量激素肌肉注射联合内毒素脂多糖(Lps)静脉注射诱导动物模型;A组注射等剂量生理盐水。分别于第2,4,6,8,10周将A、B两组各处死4只动物,取双侧股骨头标本行HE染色,测定骨髓腔内脂肪细胞面积百分比、空骨陷凹率、骨小梁面积百分比。结果:1)大剂量激素联合LPS引起B组动物股骨头内脂肪细胞肥大,第4周后脂肪细胞面积百分比大于A组(P<0.05),第6周后B组空骨陷凹率大于A组(P<0.05),第8周后B组可见骨小梁面积百分比小于A组(P<0.05)。结论:大剂量激素联合脂多糖可以很好的复制股骨头坏死模型。
OBJECTIVE: To establish an animal model of osteonecrosis of the femoral head (ONFH) by using hormone combined with lipopolysaccharide (LPS). Methods: Forty four-month-old New Zealand white rabbits were randomly divided into two groups, 20 in each group. Group A was control group and group B was model group. Group B started the first week of modeling, the use of high-dose hormone intramuscular injection of lipopolysaccharide (LPS) intravenous injection of animal model; A group was injected with normal saline. At the 2nd, 4th, 6th, 8th, 10th and 10th weeks respectively, 4 animals were sacrificed in groups A and B respectively. Bilateral femoral head specimens were examined by HE staining to determine the percentage of adipose tissue area in the marrow cavity, Trabecular area percentage. Results: (1) Hyperoxia of femoral head fat was found in group B after high dose of hormones combined with LPS. The percentage of adipocyte area in group B after 4 weeks was higher than that in group A (P <0.05). After 6 weeks, (P <0.05). The percentage of trabecular area in group B after 8 weeks was lower than that in group A (P <0.05). Conclusion: High-dose hormones combined with lipopolysaccharide can reproduce the model of femoral head necrosis well.
