目的通过结晶硫化镍在支气管上皮细胞(16HBE)中磷酸化氨基未端蛋白激酶(P-JNK)表达,探讨镍致癌的分子机制。方法用SDS聚丙烯酰胺凝胶电泳法(SDS-PAGE)检测16HBE细胞中P-JNK的表达。结果硫化镍能明显激活磷酸化氨基未端蛋白激酶(P-JNK)的表达,硫化镍浓度越高,磷酸化P-JNK的表达值越高,而且存在剂量-效应关系(r=0.7736),染镍组与对照组比较,差异有统计学意义(P<0.05)。结论结晶型硫化镍激活16HBE细胞中P-JNK的表达,可能是镍致癌的分子机制之一。
Objective To investigate the molecular mechanism of nickel carcinogenesis through the expression of phosphorylated amino-terminal kinase (P-JNK) in bronchial epithelial cells (16HBE). Methods The expression of P-JNK in 16HBE cells was detected by SDS polyacrylamide gel electrophoresis (SDS-PAGE). Results Nickel sulphide significantly activated the expression of phosphorylated amino-terminal kinase (P-JNK). The higher the concentration of nickel sulphate, the higher the expression of phosphorylated P-JNK and the dose-response relationship (r = 0.7736) Compared with the control group, the difference was statistically significant (P <0.05). Conclusion The expression of P-JNK in crystalline nickel sulfide activated 16HBE cells may be one of the molecular mechanisms of carcinogenesis of nickel.
