目的建立高效液相色谱法测定大黄虫?丸中大黄素和大黄酚的含量。方法采用Supelcosil C18色谱柱(5μm,15 cm×4.6 mm);甲醇-0.1%(ω)磷酸(体积比83∶17)为流动相;流速为1 mL/m in;柱温为30℃;检测波长为254 nm。结果大黄素在0.0196~0.2352μg/mL范围内呈良好线性关系(r=0.9999);大黄酚在0.0424~1.060μg/mL范围内呈良好线性关系(r=0.9999);平均回收率大黄素为97.5%,RSD=1.48%,大黄酚为101.5%,RSD=1.57%。结论本法准确可靠,简便迅速,适用于测定大黄虫?丸中大黄素和大黄酚的含量。
Objective To establish a HPLC method for determination of emodin and chrysophanol in Dahuangzhuang Pills. Methods Supelcosil C18 column (5μm, 15 cm×4.6 mm) was used; methanol-0.1% (ω) phosphoric acid (volume ratio 83:17) was used as mobile phase; flow rate was 1 mL/min; column temperature was 30°C; The wavelength is 254 nm. Results The emodin was linear in the range of 0.0196-0.2352μg/mL (r=0.9999); the chrysophanol was in a good linear range (0.049-1.060μg/mL) (r=0.9999); the average recovery rate of emodin was 97.5. %, RSD = 1.48%, chrysophanol 101.5%, RSD = 1.57%. Conclusion This method is accurate, reliable, simple and rapid, and is suitable for the determination of emodin and chrysophanol in Dahuangzhuang Pill.
