目的:建立同时测定玉屏风口服液中1个异黄酮类成分(毛蕊异黄酮葡萄糖苷)及3个色原酮类成分(升麻素苷、升麻素、5-O-甲基维斯阿米醇苷)含量的高效液相色谱法。方法:采用Diamonsil C18(4.6 mm×250 mm,5μm)色谱柱,以乙腈-水(17∶83)为流动相,流速1.0 m L·min~(-1),检测波长为254 nm。结果:毛蕊异黄酮葡萄糖苷、升麻素苷、升麻素和5-O-甲基维斯阿米醇苷进样量分别为0.127~2.536μg(r=0.999 9)、0.097~1.944μg(r=0.999 9)、0.081~1.616μg(r=0.999 9)和0.202~4.040μg(r=0.999 9)范围内与峰面积呈现良好的线性关系;平均回收率(n=6)分别为99.8%、99.2%、99.9%和100.5%,RSD分别为1.0%、1.2%、1.0%和1.0%。测得毛蕊异黄酮葡萄糖苷、升麻素苷、升麻素和5-O-甲基维斯阿米醇苷的含量分别为0.100~0.101、0.087~0.089、0.074~0.075和0.183~0.185 mg·m L~(-1)。结论:经方法学验证,本文建立的方法可用于玉屏风口服液的质量控制。
OBJECTIVE: To establish a method for the simultaneous determination of one isoflavone component (calycosin glucoside) and three chromogenones (cinnarinoside, cilnidin, 5-O- Alcohol glycosides) content of high performance liquid chromatography. Methods: Diamonsil C18 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase consisted of acetonitrile and water (17:83) with a flow rate of 1.0 mL · min -1 and a detection wavelength of 254 nm. Results: The injection rates of calycosin, cimicifuga, cimicifuga and 5-O-methyl-vinblastin were 0.127-2.536μg (r = 0.999 9) and 0.097-1.944μg = 0.999 9), 0.081-1.616 μg (r = 0.999 9) and 0.202-4.040 μg (r = 0.999 9). The average recoveries (n = 6) were 99.8% 99.2%, 99.9% and 100.5% with RSDs of 1.0%, 1.2%, 1.0% and 1.0%, respectively. The contents of calycosin, cimicifuga, cimicifuga and 5-O-methyl-vinblastin were found to be 0.100-0.101, 0.087-0.089, 0.074-0.075 and 0.183-0.18 mg · m L ~ (-1). Conclusion: The method established in this paper can be used for the quality control of Yupingfeng Oral Liquid.
