目的:探讨 S V40 早期基因编码产物大 T 抗原( Tag) 表达及与抑癌蛋白p Rb 的相互作用在人脑肿瘤发生发展中的意义。方法:采用免疫共沉淀及 Western 印迹法检测43 例人脑肿瘤组织及5 例正常人脑组织中 Tag 的表达,并对15 例 Tag 阳性瘤组织检测 Tag - p Rb 复合物的存在。结果: Tag 在5 例室管膜瘤及2 例脉络丛乳头状瘤中全部表达,垂体腺瘤(5/6) 、星形胶质细胞瘤(7/10) 、脑膜瘤(4/6) 、多形性胶质母细胞瘤(2/5) 及髓母细胞瘤(1/5) 均有 Tag 的表达; 3 例少枝胶质细胞瘤、1 例松果体瘤及5 例正常人脑组织无 Tag 表达。15 例 Tag 阳性瘤组织中均发现 Tag 与p Rb 形成特异性复合物。结论:在人脑肿瘤组织中 Tag 广泛表达, Tag 可与p Rb 形成特异性复合物, Tagp Rb特异性复合物的形成导致p Rb 失活,可能是人脑肿瘤发生发展的一个重要机理。
OBJECTIVE: To investigate the significance of the interaction between the expression of large T-tag (T-tag) and the tumor suppressor protein pRb in the early stage of SV40 gene in the development of human brain tumors. METHODS: The expression of Tag in 43 cases of human brain tumor tissues and 5 cases of normal human brain tissues was detected by co-immunoprecipitation and Western blot, and the presence of the Tag-p Rb complex was detected in 15 cases of Tag-positive tumor tissues. Results: Tag was expressed in 5 cases of ependymoma and 2 cases of choroid plexus papilloma. Pituitary adenoma (5/6), astrocytoma (7/10), and meningioma (4/6) , pleomorphic glioblastoma (2/5) and medulloblastoma (1/5) all had Tag expression; 3 cases of oligodendroglioma, 1 case of pineal tumor and 5 cases of normal Brain tissue without Tag expression. A specific complex of Tag and pRb was found in 15 cases of Tag-positive tumor tissues. Conclusion: Tag is widely expressed in human brain tumor tissues, and Tag can form a specific complex with p Rb. The formation of a Tagp Rb specific complex leads to the inactivation of p Rb, which may be an important mechanism for the development of human brain tumors. .
