目的:探讨牛心柿叶多糖对链脲佐菌素致糖尿病小鼠肾脏损伤的保护作用及其机制。方法:建立糖尿病小鼠模型,随机分成5组:模型组、二甲双胍阳性组及牛心柿叶多糖低、中、高剂量组。ig给予牛心柿叶多糖0.3,0.6,1.2 g·kg-1,阳性组给予二甲双胍0.32 g·kg-1,连续ig 15 d。末次给药后,氧化酶法测定空腹血糖(FBG),终点法测24 h尿微量白蛋白含量。苏木精-伊红染色法(HE染色)观察肾脏形态学变化。蛋白质印迹(Western blot)法检测肾脏组织中胰岛素样生长因子(IGF-1)表达。结果:与模型对照组比较,牛心柿叶多糖给药组中糖尿病小鼠的血糖明显降低(P<0.05),并显著降低尿微量白蛋白含量(P<0.05)。下调肾脏组织中IGF-1蛋白水平(P<0.05),并缓解肾损伤程度。结论:牛心柿叶多糖对链脲佐菌素所致糖尿病小鼠肾脏损害具有保护作用,机制可能与其通过抑制肾脏组织中IGF-1的表达而调节其下游通路有关。
Objective: To investigate the protective effect of polysaccharides from Pulsatilla chinensis on renal injury induced by streptozotocin in diabetic mice and its mechanism. Methods: Diabetic mouse models were established and randomly divided into 5 groups: model group, metformin-positive group and polysaccharide of pullulan, low, medium and high dose groups. ig given 0.3, 0.6, 1.2 g · kg-1 of polysaccharides, and the positive group was given metformin 0.32 g · kg-1 for 15 days. After the last administration, fasting blood glucose (FBG) was measured by oxidase method, and urine microalbuminuria was measured by end point method. Morphological changes of kidney were observed by hematoxylin-eosin staining (HE staining). The expression of insulin-like growth factor (IGF-1) in kidneys was detected by Western blot. Results: Compared with the model control group, the blood glucose of diabetes mellitus group was significantly decreased (P <0.05) and the urine microalbumin content was significantly decreased (P <0.05). The level of IGF-1 in kidney tissue was decreased (P <0.05) and the degree of renal injury was relieved. CONCLUSION: Polysaccharide from S. persicae has a protective effect on kidney damage induced by streptozotocin in diabetic mice, and its mechanism may be related to its regulation of its downstream pathway by inhibiting the expression of IGF-1 in kidney.
