目的:观察不同用量益气药配伍活血药对不同时间小鼠Lewis肺癌生长和转移的影响。方法:观察14和21d各组合计140只荷瘤小鼠肺转移抑制率和瘤质量,流式细胞术检测小鼠脾CD4+T淋巴细胞Th17/Treg细胞百分比动态变化。结果:各组荷瘤小鼠肺转移情况(荷瘤对照组100.00%,单纯活血药组80.27%,环磷酰胺组76.39%,益气活血高、中和低剂量组均为68.55%)、14d瘤质量〔各组分别为(1.19±0.13)、(1.17±0.09)、(0.79±0.07)、(1.43±0.10)、(1.38±0.30)和(0.86±0.09)g〕、14dTh17〔各组分别为(0.99±0.13)%、(0.86±0.09)%、(0.79±0.07)%、(0.78±0.10)%、(0.78±0.30)%和(0.77±0.09)%〕和14dTreg〔各组分别为(9.44±0.91)%、(8.12±0.34)%、(7.12±0.66)%、(7.00±0.14)%、(7.01±0.12)%和(7.03±0.12)%〕细胞百分比及比值动态变化比较,其余各组较荷瘤对照组差异均有统计学意义,P<0.05。各益气活血药组荷瘤小鼠肺转移情况、Th17和Treg细胞百分比及比值动态变化均优于单纯活血药组,P<0.05;而与环磷酰胺组相似。各治疗组瘤质量以益气活血药高剂量组最重,其次为益气活血药中剂量组,两者高于荷瘤对照组,与其余各组比较差异有统计学意义,P<0.05,但益气活血小剂量组优于高、中剂量组,而与环磷酰胺组相似。各组肿瘤细胞增殖与瘤质量呈现出相一致情况,而肿瘤细胞凋亡与瘤质量基本呈现出相反情况。结论:益气活血药物较单纯活血药,对肿瘤转移有较好的抑制作用,但早期妄用益气药有助邪之害而促使肿瘤生长,小剂量的益气药配伍活血药,应是不同病程时期控制肿瘤转移和生长的稳妥方案;同时得出,肿瘤生长和转移间不存在机制上的必然联系。
Objective: To observe the effects of different dosage of Qi-supplementing herbs and promoting blood circulation on the growth and metastasis of Lewis lung carcinoma in different time. Methods: The inhibitory rate of lung metastasis and the tumor mass of 140 tumor-bearing mice in each group were observed on the 14th and 21th day. The percentage of Th17 / Treg cells in splenic CD4 + T lymphocytes was detected by flow cytometry. The remaining groups than the tumor control group differences were statistically significant, P <0.05. The lung metastasis, the percentage of Th17 and Treg cells and the ratio of dynamic changes in the Tumor-bearing mice of Yiqi Huoxue Decoction group were all better than those of Huoxue Huoxue Decoction group (P <0.05), but similar to cyclophosphamide group. The tumor weight of each treatment group was the highest in the high-dose Yiqi Huoxue medicine group, followed by the middle-dose Yiqihuoxue medicine group, both of which were higher than that of the tumor-bearing control group, with statistical significance (P <0.05) However, the Yiqi Huoxue low-dose group is superior to the high and middle dose groups, but similar to the cyclophosphamide group. The proliferation of tumor cells in each group showed consistent with the tumor mass, while the apoptosis of tumor cells and tumor mass showed the opposite situation. Conclusion: Yiqihuoxue drugs have better inhibitory effect on tumor metastasis than pure blood-activating drugs, but the early use of Yiqi drug can help the evil and promote the growth of the tumor.
