本实验采用皮下递增注射吗啡5 天后,经纳络酮催瘾建立吗啡戒断模型。观察中枢5HT1A受体激动剂乌拉地尔(urapidil) 对吗啡依赖大鼠戒断后的平均动脉压(mABP) 和心率(HR) 的影响。实验结果表明,侧脑室微量注射乌拉地尔具有明显抑制吗啡戒断后血压升高的作用,并呈量效关系。乌拉地尔对吗啡戒断后心率加速虽也有抑制作用,但不呈量效关系。用另一种特异性5HT1 A受体激动剂8OHDPAT兴奋中枢5HT1A受体时也得到相同的结果。
In this experiment, morphine withdrawal model was established by naloxone withdrawal after increasing subcutaneous injection of morphine for 5 days. To observe the effect of 5-HT1A receptor agonist urapidil on the mean arterial pressure (mABP) and heart rate (HR) after withdrawal from morphine-dependent rats. The experimental results show that microinjection of urapidilazine into the lateral ventricle significantly inhibits the increase of blood pressure after morphine withdrawal, and has a dose-response relationship. Urapidil also had an inhibitory effect on the acceleration of heart rate after withdrawal from morphine but did not show a dose-response relationship. The same result was obtained with another specific 5-HT1 A receptor agonist 8-OH DPAT excitement center 5 HT1A receptor.
