目的通过癃畅颗粒对大鼠前列腺增生组织凋亡调控基因fas表达的影响,探讨癃畅颗粒治疗前列腺增生的作用机制。方法 Wistar雄性大鼠120只,随机平均分为癃畅颗粒高、中、低剂量组,癃闭舒组,模型组和正常组6组,每组20只。除正常组外,其余各组采用大鼠去势后注射Testosterone Propionate(丙酸睾酮)至前列腺增生法造模,各组均灌胃给药30d后处死,摘取前列腺组织测量湿质量,计算前列腺指数,采用免疫组化PV法检测实验鼠前列腺增生组织Fas表达状况。结果(1)前列腺湿重:正常组(0.62±0.91)g;模型组(0.92±0.50)g;癃闭舒组(0.75±0.58)g;癃畅颗粒低剂量组(0.80±0.92)g;癃畅颗粒中剂量组(0.79±0.10)g;癃畅颗粒高剂量组(0.64±0.71)g;与模型组比较,差异有统计学意义(P<0.05)。(2)前列腺指数:正常组(0.142±0.061),模型组(0.225±0.064),癃闭舒组(0.168±0.092),癃畅颗粒低剂量组(0.199±0.047),癃畅颗粒中剂量组(0.182±0.036),癃畅颗粒高剂量组(0.167±0.039),与模型组比较,差异有统计学意义(P<0.05)。(3)癃畅颗粒对良性前列腺增生大鼠前列腺上皮细胞fas基因表达影响(平均光密度):正常组(0.216±0.029),模型组(0.183±0.089),癃闭舒组(0.201±0.129),癃畅颗粒低剂量组(0.200±0.126),癃畅颗粒中剂量组(0.202±0.143),癃畅颗粒高剂量组(0.207±0.177),与模型组比较,差异有统计学意义(P<0.05)。结论癃畅颗粒能够上调大鼠前列腺组织fas表达,缩小模型大鼠前列腺湿质量,减轻病理变化,这可能是该药临床有效治疗前列腺增生症的作用机制之一。
Objective To study the effect of Bingchang granule on the expression of fas in rats with benign prostatic hyperplasia and to explore the mechanism of Changchang granule in treating benign prostatic hyperplasia. Methods A total of 120 Wistar male rats were randomly divided into 6 groups (n = 20), including high, medium and low doses of Cimici Granules, Zu Shutushu group, model group and normal group. Except for the normal group, rats in other groups were injected with Testosterone Propionate (BPA) to BPH after castration in rats. The rats in each group were sacrificed 30 days after gavage administration, and the prostate tissues were harvested to measure the wet weight, and the prostate Index, using immunohistochemical PV method to detect the expression of Fas in experimental benign prostatic hyperplasia. Results: (1) Wet weight of the prostatic: normal group (0.62 ± 0.91) g; model group (0.92 ± 0.50) g; (0.79 ± 0.10) g and Pian Chang granules high-dose group (0.64 ± 0.71) g. Compared with the model group, the difference was statistically significant (P <0.05). (2) Prostate index: normal group (0.142 ± 0.061), model group (0.225 ± 0.064), 癃 closed Shu group (0.168 ± 0.092), 癃 Chang particle low dose group (0.199 ± 0.047) (0.182 ± 0.036) and Changzhi Granules high dose group (0.167 ± 0.039), respectively. The difference was statistically significant (P <0.05) compared with model group. (3) The effect of Chengchang granule on the fas gene expression in benign prostatic hyperplasia rat prostate epithelial cells (average optical density): 0.216 ± 0.029 in the normal group, 0.183 ± 0.089 in the model group, 0.201 ± 0.129 in the control group, (0.200 ± 0.126), Zhongchang Granule medium dose group (0.202 ± 0.143) and Changchang Granule high dose group (0.207 ± 0.177), the difference was statistically significant (P < 0.05). Conclusion Cheng Chang granules can upregulate the expression of fas in rat prostate tissue and reduce the wet mass of prostate in model rats, alleviating the pathological changes, which may be one of the effective mechanisms of this drug in the treatment of benign prostatic hyperplasia.
