目的:通过对火绒草不同提取物处理高血糖小鼠血糖的测定,探讨火绒草提取物降血糖的有效部位和剂量。方法:火绒草用70%乙醇回流提取,提取物上D101大孔树脂柱,依次用50%,70%,95%的乙醇洗脱,得到50%,70%,95%的乙醇提取物。采用链脲佐菌素(STZ)诱导高血糖小鼠,7 d后分别用二甲双胍(300 mg·kg-1),50%乙醇提取物(100 mg·kg-1),70%乙醇提取物(100 mg·kg-1)和95%乙醇提取物(100 mg·kg-1)ig给药,模型组和空白组以等量的蒸馏水代替给药,10 d后测定高血糖小鼠的空腹血糖。将筛选出的有效降糖部位分为高、中、低(400,200,100 mg·kg-1)剂量处理高血糖小鼠,10 d后测定空腹血糖。结果:与空白组比较,模型组血糖显著升高(P<0.01)。与模型组比较,火绒草的不同提取物均能降低高血糖小鼠血糖,50%乙醇提取物具有显著的降血糖效果,且以中剂量为佳(P<0.01)。结论:火绒草的降血糖有效部位为50%乙醇提取物。
OBJECTIVE: To investigate the effect of different extracts of Tetromorpha on the determination of blood glucose in mice with hyperglycemia, and to explore the effective sites and dosages of the extract. Methods: The extract was extracted with 70% ethanol. The extract was applied to D101 macroporous resin column and eluted with 50%, 70% and 95% ethanol to obtain 50%, 70% and 95% ethanol extract. High glucose mice were induced by streptozotocin (STZ) and were treated with metformin (300 mg · kg -1), 50% ethanol (100 mg · kg -1), 70% ethanol extract 100 mg · kg-1) and 95% ethanol extract (100 mg · kg-1). The rats in the model group and the blank group were treated with equal volume of distilled water. After 10 days, the fasting blood glucose . The selected hypoglycemic sites were divided into high, medium and low (400,200,100 mg · kg-1) doses of hyperglycemic mice, fasting blood glucose was measured after 10 days. Results: Compared with the blank group, the model group, blood glucose was significantly increased (P <0.01). Compared with the model group, different extract of Erigeron could reduce blood glucose in mice with hyperglycemia, 50% ethanol extract had significant hypoglycemic effect, and the best dose (P <0.01). Conclusion: The bletilla striata glycosides hypoglycemic effective site is 50% ethanol extract.
