目的:探讨顺铂对卵巢癌细胞凋亡的诱导作用,以揭示顺铂治疗卵巢癌的作用机理。方法:选用不同剂量顺铂与人卵巢癌细胞系HO-8910共同孵育不同时间后,应用光镜、电镜进行形态学观察。提取细胞DNA经琼脂糖凝胶电泳分析电泳带型。细胞DNA经特异性荧光染色后,用流式细胞仪分析细胞周期变化。结果:细胞经顺铂处理后浓缩、深染,致密的染色质沿核膜下凝聚,有凋亡小体形成。细胞DNA电泳呈现梯状带型。细胞周期分析G1峰前出现明显的细胞凋亡峰,以25mg/L顺铂作用3h、50mg/L作用1h为例,定量分析凋亡细胞发生率分别为15.4%和44.5%。结论:顺铂可通过诱导人卵巢癌细胞系HO-8910发生细胞凋亡达到消灭肿瘤细胞的目的。
Objective: To investigate the inducing effect of cisplatin on apoptosis of ovarian cancer cells in order to reveal the mechanism of action of cisplatin on ovarian cancer. METHODS: Different doses of cisplatin were incubated with human ovarian cancer cell line HO-8910 for different periods of time. Light microscope and electron microscope were used for morphological observation. Cellular DNA was extracted and analyzed by agarose gel electrophoresis. Cellular DNA was stained with specific fluorescence, and changes in cell cycle were analyzed by flow cytometry. RESULTS: After cisplatin treatment, the cells were concentrated and deeply stained. The dense chromatin condensed along the nuclear membrane and formed apoptotic bodies. Cell DNA electrophoresis presents a ladder pattern. Apoptotic peaks appeared before the G1 peak in cell cycle analysis. Taking 25mg/L cisplatin for 3h and 50mg/L for 1h as an example, the incidence of apoptotic cells was quantified by 15.4% and 44.5%, respectively. CONCLUSION: Cisplatin can eliminate tumor cells by inducing apoptosis in HO-8910 ovarian cancer cell line.
