目的验证不同浓度的辛伐他汀对人类成骨细胞(OB)功能表达和体外成骨能力的影响,研究雌激素促进成骨作用的机制及剂量效应关系。方法取成人的髂骨松质骨,采用胶原-胰蛋白酶消化法,获得松质骨中的成骨细胞,进行纯化和培养。在此基础上添加不同浓度的辛伐他汀(1×10-10, 1×10-9, 1×10-8, 1×10-7, 1×10-6, 1×5-5 mol/L)并进行培养。用生化法测定细胞碱性磷酸酶(ALP)活性,用放射免疫法测定细胞培养液中骨钙素(OC)和细胞间质的钙含量。结果辛伐他汀对成骨细胞ALP活性和骨钙素分泌的影响均呈正相关,即各浓度辛伐他汀对ALP活性、骨钙素的分泌和成骨细胞的成骨能力均有刺激作用,并与辛伐他汀剂量呈正相关。结论辛伐他汀能增加成骨细胞的ALP活性和骨钙素的产生,提高成骨细胞的成骨能力。
Objective To verify the effect of different concentrations of simvastatin on the functional expression of human osteoblasts (OB) and the osteogenic ability in vitro and to study the mechanism of estrogen in promoting osteogenesis and the dose-response relationship. Methods Adult iliac cancellous bone was obtained. The osteoblasts in cancellous bone were obtained by collagen-trypsin digestion and purified. On this basis, different concentrations of simvastatin (1 × 10-10, 1 × 10-9, 1 × 10-8, 1 × 10-7, 1 × 10-6, 1 × 5-5 mol / L ) And cultured. The alkaline phosphatase (ALP) activity of cells was determined by biochemical method, and the content of osteocalcin (OC) and intercellular calcium in cell culture medium were determined by radioimmunoassay. Results Simvastatin had a positive correlation with ALP activity and osteocalcin secretion in osteoblasts. Simvastatin at various concentrations had a stimulatory effect on ALP activity, secretion of osteocalcin and osteogenic ability of osteoblasts There was a positive correlation with the dose of simvastatin. Conclusion Simvastatin can increase osteoblast ALP activity and osteocalcin production and increase osteoblast osteogenic capacity.
