目的:建立HPLC法测定痤疮清软膏经皮给药后大鼠体内中甲硝唑含量。方法:采用反相高效液相色谱法,Phenomsil C18柱(250mm×4.6mm,5μm)色谱柱;流动相:甲醇-水(10:90);检测波长:277nm;柱温:30℃;进样量为20μL;流速:1.0mL.min-1。结果:方法检出限为0.212μg/mL;甲硝唑在0.212~6.784μg/mL范围内线性良好,r=0.9991;样品加标回收率为96.2%,RSD=3.5。结论:该方法准确、稳定,有良好的重现性和较高的灵敏度。
OBJECTIVE: To establish an HPLC method for the determination of metronidazole in rats after transdermal administration of acne clearing ointment. Methods: The mobile phase consisted of methanol-water (10: 90), detection wavelength of 277 nm and column temperature of 30 ℃. The sample was separated on a Phenomicil C18 column (250 mm × 4.6 mm, 5 μm) using reversed-phase high performance liquid chromatography Amount was 20 μL; flow rate: 1.0 mL.min-1. Results: The detection limit of this method was 0.212 μg / mL. Metronidazole was linear in the range of 0.212 ~ 6.784 μg / mL with r = 0.9991. The recoveries of spiked samples were 96.2% with RSD of 3.5. Conclusion: The method is accurate, stable, good reproducibility and high sensitivity.
