目的 探讨Fos和Jun的表达产物与人肝细胞癌发生、发展的关系。方法 采用免疫组织化学S P法和蛋白电泳技术对 2 0例肝癌病人癌及癌旁组织中Fos和Jun核内癌蛋白的表达情况进行检测。结果 Westernblot结果示 2 0例标本中有 16例肝癌组织FosB、JunD蛋白含量高于癌旁组织。FosB光密度值在肝癌及癌旁组织中分别为 2 396± 80 3和 894± 319,差异有显著意义 (P <0 0 1) ;JunD光密度值在肝癌及癌旁组织中分别为 1973± 72 5和 776± 16 1,差异有显著意义 (P <0 0 5 )。免疫组织化学结果显示FosB和JunD蛋白在肝癌组织细胞核中同步表达。结论 在人原发性肝细胞癌组织中 ,核内转录基因FosB、JunD表达产物表达强度明显增高 ,这可能是肝细胞早期恶变的重要原因之一。
Objective To investigate the relationship between the expression products of Fos and Jun and the occurrence and development of human hepatocellular carcinoma. Methods Immunohistochemistry SP method and protein electrophoresis technique were used to detect the expression of Fos and Jun nuclear oncoproteins in cancer tissues and paracancerous tissues of 20 patients with liver cancer. Results The results of Westernblot showed that the contents of FosB and JunD proteins were higher in 16 cases of HCC than in adjacent tissues. The optical density of FosB was 2 396± 80 3 and 894± 319 in hepatocellular carcinoma and adjacent tissues, respectively (P <0 01). JunD optical density was 1973 in hepatocellular carcinoma and adjacent tissues. 72 5 and 776 ± 16 1, the difference was significant (P <0 05). Immunohistochemistry results showed that FosB and JunD proteins were synchronously expressed in the nucleus of HCC tissues. Conclusion In human primary hepatocellular carcinoma, the expression of FosB and JunD transcripts is significantly increased, which may be one of the important reasons for early hepatocyte malignancy.
