背景:研究发现人源羊水干细胞可向心肌前体细胞分化。目的:观察不同浓度重组人促红细胞生成素对人源羊水干细胞向心肌前体细胞分化的影响。方法:用含0,1.0,5.0,10.0,20.0U/mL重组人促红细胞生成素的诱导培养基诱导鉴定后的人源羊水干细胞向心肌前体细胞分化,诱导24h后换为完全培养基继续培养。结果与结论:诱导分化14d,倒置显微镜下可见细胞由长梭形逐渐变短增宽,相邻的细胞间有融合现象,似类肌管样结构,以5.0U/mL重组人促红细胞生成素的诱导分化率最高;RT-PCR检测显示重组人促红细胞生成素可促进细胞Nkx-2.5和GATA-4mRNA的表达,以5.0U/mL重组人促红细胞生成素的作用最强。说明外源性重组人促红细胞生成素能剂量依赖性促进人源羊水干细胞向心肌前体细胞分化。
Background: The study found that human amniotic fluid stem cells differentiate into cardiomyocyte precursors. Objective: To observe the effect of different concentrations of recombinant human erythropoietin on the differentiation of human amniotic fluid stem cells into cardiomyocyte progenitor cells. Methods: Induced differentiation of human amniotic fluid derived stem cells into cardiomyocyte progenitor cells with induction medium containing 0, 1.0, 5.0, 10.0, 20.0 U / mL recombinant human erythropoietin, and then induced to complete medium for 24 h to cultivate. RESULTS AND CONCLUSION: After differentiation for 14 days, under inverted microscope, the cells gradually became shorter and broadened by the long fusiform shape, and the adjacent cells had the phenomenon of fusion, similar to the myotube-like structure. The 5.0U / mL recombinant human erythropoietin RT-PCR assay showed that recombinant human erythropoietin can promote the expression of Nkx-2.5 and GATA-4 mRNA, and the effect of 5.0U / mL recombinant human erythropoietin was the strongest. It shows that exogenous recombinant human erythropoietin can promote the differentiation of human amniotic fluid stem cells into cardiomyocyte progenitor cells in a dose-dependent manner.
