作者应用逆转录-多聚酶链反应(RT-PCR)检测25例急性白血病患者骨髓细胞中mdr_1基因的表达。以K562敏感细胞和体外诱导的耐药细胞株K562/A02分别作为阴性和阳性对照,发现复发和未缓解患者中mdr_1表达增高者占90.9%(10/11),而初治患者中仅为21.4%(3/14),mdr_1表达增高者化疗效果差。应用单克隆抗体JSB-1检测P-170结果与RT-PCR一致。此RT-PCR具有高度敏感性,可以检出2Pg的mdr_1cDNA,对于检测白血病mdr_1的表达是一种灵敏、特异的方法。
The authors used reverse transcription-polymerase chain reaction (RT-PCR) to detect the expression of mdr1 gene in bone marrow cells of 25 patients with acute leukemia. The K562-sensitive cells and the in vitro induced drug-resistant cell line K562/A02 were used as negative and positive controls, respectively, and 90.9% (10/11) of the patients with relapsed and unresponsive mdr-1 expression were found to increase. However, in the untreated patients, only It was 21.4% (3/14), and mdr_1 expression increased in patients with poor chemotherapy. The detection of P-170 using monoclonal antibody JSB-1 was consistent with RT-PCR. This RT-PCR is highly sensitive and can detect 2Pg of mdr_1 cDNA, which is a sensitive and specific method for detecting the expression of mdr_1 in leukemia.
