钙调神经磷酸酶信号转导通路在结直肠癌组织中的表达

目的检测钙调神经磷酸酶(CaN)信号转导通路在结直肠癌组织中的表达,分析CaN成员与结直肠癌临床病理特征的关系。方法应用Western blot检测40例结直肠癌组织及其邻近正常黏膜中CaN Aa、CaN Aβ、CaN B与bcl-2蛋白表达,免疫组织化学法检测CaN在细胞水平的分布。结果结直肠癌组织中CaN信号传导通路成员CaN Aa、CaN Aβ、CaN B与bcl-2在结直肠癌组织中表达明显升高,分别为正常黏膜的3.45倍、3.67倍、4.31倍与3.18倍(P<0.05);CaN Aα与CaN Aβ表达水平与TNM分期较晚(Ⅲ/Ⅳ)有关(P<0.05),CaN Aα与bcl-2在结直肠癌组织中表达情况呈线性相关,相关系数为0.493,P<0.05。结论CaN信号转导通路持续活化可能在结直肠癌的发生中起重要作用。 Objective To detect the expression of calcineurin (CaN) signal transduction pathway in colorectal cancer tissues and to analyze the relationship between CaN members and the clinicopathological features of colorectal cancer. Methods Western blot was used to detect the protein expressions of CaN Aa, CaN Aβ, CaN B and bcl-2 in 40 cases of colorectal cancer tissue and adjacent normal mucosa. The distribution of CaN at the cellular level was detected by immunohistochemistry. Results The expressions of CaN Aa, CaN Aβ, CaN B and bcl-2 in colorectal cancer tissues were significantly increased in colorectal cancer tissues, which were 3.45 folds, 3.67 folds, and 4.67 folds higher than that of normal mucosa .31 and 3.18 respectively (P <0.05). The expressions of CaN Aα and CaN Aβ were correlated with the later stages of TNM (Ⅲ / Ⅳ) (P <0.05). The expressions of CaN Aα and bcl- Rectal cancer tissue expression was linearly correlated, the correlation coefficient was 0.493, P <0.05. Conclusion The continuous activation of CaN signal transduction pathway may play an important role in the development of colorectal cancer.
