用人肺腺癌细胞株AGZ—83a常规免疫BALB/C小鼠,取脾细胞与小鼠骨髓瘤细胞SP2/0融合,获得一稳定分泌抗人肺腺癌单克隆抗体(McAb)的杂交癌细胞林──—QMC-WL99。制备并纯化了小鼠腹水抗体,腹水效价为5.5X106,亚型鉴定为IgG1。免疫组化结果显示QMC-WL99单抗的特异性较高,与肺腺癌有较好选择反应性,与正常组织无交及反应。该单克隆抗体的亲和常数为7.25×1010M-1,液氮冻再后复苏,杂交瘤细胞株QMC-WL99分泌抗体的能力不受影响。
BALB/C mice were routinely immunized with human lung adenocarcinoma cell line AGZ-83a, and spleen cells were fused with mouse myeloma cells SP2/0 to obtain a hybrid cancer cell that stably secreted anti-human lung adenocarcinoma monoclonal antibody (McAb). Lin --- QMC-WL99. The mouse ascites antibody was prepared and purified. The titer in ascites was 5.5X106 and the subtype was identified as IgG1. The results of immunohistochemistry showed that the specificity of QMC-WL99 mAb was higher, and it had a better selective response to lung adenocarcinoma, and no reaction with normal tissues. The affinity constant of the monoclonal antibody was 7.25×1010M-1. After the liquid nitrogen was frozen and then revived, the ability of the hybridoma cell strain QMC-WL99 to secrete antibodies was not affected.
