目的研究贝伐单抗对机械通气引起的肺损伤的预防作用。方法 18只Wistar大鼠随机均分为三组:假手术(A)组、机械通气(B)组和贝伐单抗20mg/kg治疗(C)组。通气结束后,常规取肺泡灌洗液与肺组织,ELISA法检测灌洗液中的血管内皮生长因子(VEGF)浓度,Brodfard法测定蛋白含量,依文氏蓝含量和肺干/湿重比值评价肺毛细血管通透性,HE染色观察肺组织的病理改变并计算肺损伤指数。结果与A组相比,B组肺泡灌洗液中VEGF浓度、肺毛细血管通透性与肺损伤均显著增加(P<0.05),而C组上述指标均较B组明显降低(P<0.05)。结论贝伐单抗对机械通气引起的肺损伤有一定的预防效应。
Objective To study the preventive effect of bevacizumab on lung injury induced by mechanical ventilation. Methods Eighteen Wistar rats were randomly divided into three groups: sham operation (A), mechanical ventilation (B) and bevacizumab 20mg / kg (C). After ventilation, alveolar lavage fluid and lung tissue were routinely collected. The concentration of vascular endothelial growth factor (VEGF) in lavage fluid was detected by ELISA, the protein content, the content of Evans blue and the ratio of lung dry weight / wet weight Pulmonary capillary permeability, HE staining observed pathological changes of lung tissue and calculate the lung injury index. Results Compared with group A, VEGF concentration, pulmonary capillary permeability and lung injury in group B were significantly increased (P <0.05), while those in group C were significantly lower than those in group B (P <0.05 ). Conclusion Bevacizumab has some preventive effects on lung injury induced by mechanical ventilation.
