目的探讨二冬膏含药血清对人肺癌A549细胞的抑制效应。方法采用中药血清药理学方法,选择高、中、低剂量的二冬膏水溶液以灌胃方式给药,获取二冬膏含药血清,以空白血清为对照,干预A549细胞,应用MTT法、划痕实验法及Transwell侵袭实验法检测二冬膏含药血清对A549细胞增殖、迁移及侵袭能力的影响。结果二冬膏高、中、低剂量组干预A549细胞24 h后均可不同程度抑制肺腺癌A549细胞增殖,差异有统计学意义(P<0.05),并呈时间、剂量依赖性,以二冬膏中剂量组抑制作用最明显;二冬膏高、中、低剂量组A549细胞的迁移距离较空白组缩短,差异有统计学意义(P<0.05),中剂量组细胞迁移距离最短;二冬膏高、中、低剂量组均能降低A549细胞的侵袭能力,差异有统计学意义(P<0.05),中剂量组侵袭过膜的细胞数最少。结论二冬膏含药血清能抑制肺癌A549细胞增殖、迁移及侵袭能力,中剂量抑制作用最明显。
Objective To investigate the inhibitory effect of Erdong cream containing serum on human lung cancer A549 cells. Methods Serum pharmacology of traditional Chinese medicine (TCM) was used to select the high, medium and low doses of Dongshang cream aqueous solution for intragastric administration. The serum containing Erzhi cream was obtained. A549 cells were treated with blank serum as control. Trace assay and Transwell invasion assay were used to detect the effects of serum containing Dongnong paste on the proliferation, migration and invasion of A549 cells. Results Two-year high and medium dose Dongzhi A549 cells could inhibit the proliferation of A549 cells in different degrees after being treated for 24 h, the difference was statistically significant (P <0.05), and in a time and dose-dependent manner, The effect of Dongzao medium dose group was the most obvious. The migration distance of A549 cells in high, medium and low dose group of Dongsong paste was shorter than blank group, the difference was statistically significant (P <0.05) The high, medium and low dose of Dong Gao could reduce the invasiveness of A549 cells, the difference was statistically significant (P <0.05), and the medium dose group had the least number of cell invasion. Conclusion Two winter serum containing serum can inhibit lung cancer A549 cell proliferation, migration and invasion ability, the most obvious dose inhibition.
