目的探讨雌激素对大鼠慢性颞叶癫发作的影响及其与瞬时外向钾通道(kv4.2)之间的关系。方法 SD雌性大鼠随机分为正常对照组、去势对照组、正常致组、去势致组。应用毛果芸香碱诱导大鼠癫持续状态(SE),观察大鼠行为学变化。经4周后成为慢性颞叶癫大鼠。Western bolt方法检测大鼠海马瞬时外向钾通道Kv4.2蛋白表达水平。结果与正常致组比较,去势致组达到SE的潜伏期延长、死亡率降低,但差异无统计学意义(P>0.05);去势致组的癫发作强度较正常致组轻,差异有统计学意义(P<0.05)。去势致组和正常致组在SE 4周后海马Kv4.2均较正常对照组显著升高,均差异有统计学意义(P<0.05和P<0.01)。去势对照组和去势致组比较,均未见对Kv4.2蛋白表达产生影响,说明Kv4.2蛋白表达升高为癫发作所致。结论大鼠癫发作后Kv4.2表达增加、瞬时外向钾电流增强、神经冲动的发放频率减慢,可能是产生代偿性自我保护反应的结果;虽然去势雌性大鼠未对Kv4.2表达产生影响,但出现潜伏期延长和死亡率降低、发作强度降低的行为学趋势,提示低剂量雌激素可能存在对癫发作的保护作用。
Objective To investigate the effect of estrogen on the onset of chronic temporal lobe epilepsy in rats and its relationship with transient outward potassium channels (kv4.2). Methods SD female rats were randomly divided into normal control group, castration control group, normal induced group and castration induced group. The application of pilocarpine induced epileptic seizure state (SE) in rats, behavioral changes observed. After 4 weeks become chronic temporal lobe epileptic rats. Western bolt method was used to detect the expression level of Kv4.2 in rat hippocampal transient outward potassium channel. Results Compared with the normal group, the latent period of SE was prolonged and the mortality rate decreased in the castrated group (P> 0.05), and the intensity of epileptic seizures in the castrated group was lighter than that of the normal group Statistical significance (P <0.05). The levels of Kv4.2 in hippocampus in ovariectomized and normal induced groups were significantly higher than those in normal control group after 4 weeks of SE (P <0.05 and P <0.01). There was no effect on the expression of Kv4.2 protein in the castration control group and the castrated group, indicating that the increased expression of Kv4.2 protein was caused by the epileptic seizure. Conclusions The expression of Kv4.2 increased after transient epileptic seizures in rats, and the transient outward potassium current increased and the frequency of nerve impulses released was probably the result of compensatory self-protection reaction. Although the ovariectomized female rats did not respond to Kv4.2 Expression, but there is a trend of behavioral tendency with prolonged latency and decreased mortality and decreased seizure intensity, suggesting that low doses of estrogen may have a protective effect on epileptic seizures.
