目的:建立测定大鼠血浆中去铁酮(deferiprone,DFP)的高效液相色谱-紫外检测法。方法:采用Diamonsil(R)钻石C18色谱柱(250mm×4.6mm,5μm),流动相为乙腈-(0.05mol.L-1磷酸氢二钠、5mmol.L-1庚烷磺酸钠和2mmol.L-1ED-TA)(磷酸调pH=3.0)(8:92,v/v),流速1.0mL.min-1,紫外检测波长为278nm。结果:去铁酮浓度在1~120mg.L-1范围内,线性良好(r=0.9999),最低检出限6.0μg.L-1(S/N≥3),绝对回收率大于98.7%,相对回收率为99.0%~100.6%,日内和日间精密度RSD均小于2%。结论:该方法快速、准确、灵敏,重复性好,可用于DFP血药浓度检测和药代动力学研究。
Objective: To establish a HPLC method for the determination of deferiprone (DFP) in rat plasma. METHODS: Diamonsil (R) diamond C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile- (0.05 mol·L-1 sodium phosphate dibasic, 5 mmol.L-1 heptane sodium sulfonate and 2 mmol. L-1ED-TA) (pH adjusted to 3.0 with phosphoric acid) (8:92, v / v) at a flow rate of 1.0 mL.min-1. The UV detection wavelength was 278 nm. Results: The linearity was good (r = 0.9999) and the lowest detection limit was 6.0μg.L-1 (S / N≥3) in the range of 1 ~ 120mg.L-1. The absolute recovery was more than 98.7% The relative recoveries ranged from 99.0% to 100.6%. The intra-day and inter-day RSDs were less than 2%. Conclusion: The method is rapid, accurate, sensitive and reproducible. It can be used for DFP plasma concentration determination and pharmacokinetic studies.
