目的:研究剔毒护肝方药物血清对肝星状细胞凋亡相关蛋白Bcl-2、Bax的影响。方法:传代培养大鼠肝星状细胞(HSC)系HSC-T6与不同剂量剔毒护肝方(大、中、小剂量组)共同培养48小时,应用Annexin-V-碘化丙锭染色检测剔毒护肝方对大鼠HSC凋亡的影响;免疫组化检测凋亡相关蛋白Bcl-2、Bax水平。结果:剔毒护肝方能使大鼠HSC凋亡率明显增多,同时可使凋亡抑制分子Bcl-2表达下调,促凋亡分子Bax表达增强(P<0.01)。结论:剔毒护肝方可下调Bcl-2/Bax比率,促进HSC凋亡。
Objective: To study the effect of serum of Tiaodu Hugan recipe on the apoptosis-related proteins Bcl-2 and Bax in hepatic stellate cells. METHODS: HSC-T6 cells were subcultured for 48 hours with different doses of Tiaodu Hugan Decoction (large, medium and low dose groups) and detected by Annexin-V-propidium iodide staining. Effect of Tiaodu Hugan Decoction on the apoptosis of HSCs in rats; The levels of apoptosis-related proteins Bcl-2 and Bax were detected by immunohistochemistry. RESULTS: Tidu Hugan Decoction can significantly increase the apoptosis rate of rat HSC, at the same time, it can down-regulate the expression of apoptosis inhibitor Bcl-2 and increase the expression of pro-apoptotic molecule Bax (P<0.01). Conclusion: Tiedu Hugan prescription can down-regulate Bcl-2/Bax ratio and promote HSC apoptosis.
