目的评价重组弓形虫热休克蛋白70(rTgHSP70)联合弗氏佐剂皮下免疫小鼠的抗弓形虫感染作用。方法BALB/c小鼠70只,随机分为7组,分别为PBS组、A组(佐剂+PBS)、20PA组(20μg rTgHSP70+佐剂)、40P组(40μgrTgH-SP70)、40PA组(40μg rTgHSP70+佐剂)、60PA组(60μg rTgHSP70+佐剂)及80PA组(80μg rTgHSP70+佐剂),皮下免疫,间隔2周,加强免疫2次,共免疫3次。末次免疫后14d,用2×104个RH株弓形虫速殖子/只灌胃攻击各组小鼠,攻击后30d处死,ELISA检测血清IgG和肠液sIgA;分离脑组织内弓形虫速殖子,并计数。结果在添加佐剂组中,血清IgG水平随rTgHSP70剂量的增加而升高,与PBS组和A组比较,差异有统计学意义;肠液sIgA水平60PA组最高,与20PA和40PA组比较,差异有统计学意义;与PBS组相比,A、20PA、40P、40PA、60PA和80PA组的脑组织内速殖子数分别减少了56.87%、56.46%、61.90%、48.74%、62.87%和70.44%。结论rTgHSP70联合弗氏佐剂皮下免疫小鼠可诱导一定的抗弓形虫感染作用,80μgrTgHSP70为较佳剂量。
Objective To evaluate the anti-Toxoplasma infection effect of recombinant Toxoplasma gondii heat shock protein 70 (rTgHSP70) combined with Freund’s adjuvant in mice. Methods Seventy BALB / c mice were randomly divided into 7 groups: PBS group, A group (adjuvant plus PBS), 20PA group (20μg rTgHSP70 + adjuvant), 40P group (40μg rTgH-SP70), 40PA group rTgHSP70 + adjuvant), 60PA group (60μg rTgHSP70 + adjuvant) and 80PA group (80μg rTgHSP70 + adjuvant). The mice were immunized subcutaneously with 2 weeks interval. Twenty-four days after the last immunization, mice of each group were challenged with 2 × 104 RH Toxoplasma gondii tachyzoites / gavage and sacrificed at 30 days after challenge. Serum IgG and intestinal sIgA were detected by ELISA. Toxoplasma gondii, And count. Results In the adjuvant group, serum IgG level increased with the increase of rTgHSP70 dose, which was significantly different from PBS group and A group. The sIgA level of intestinal juice group was the highest in 60PA group, compared with 20PA group and 40PA group Compared with PBS group, the tachyzoites in brain tissue of A, 20PA, 40P, 40PA, 60PA and 80PA groups decreased by 56.87%, 56.46%, 61.90%, 48.74%, 62.87% and 70.44% . Conclusion rTgHSP70 combined with Freund’s adjuvant can induce certain anti-Toxoplasma infection in mice, and 80μg rTgHSP70 is the best dose.
