目的:对山羊角提取物中氨基酸进行含量测定研究。方法:采用高效液相柱前衍生2,4-二硝基氟苯法(DNFB-HPLC)检测山羊角提取物中常规氨基酸含量。色谱柱为Elite AAK(250 mm×4.6 mm,5μm),流动相A:乙腈-水(1∶1),流动相B:0.05 mol/L的醋酸钠缓冲液(pH=6.4),流速为1.2 mL/min,检测波长360 nm,柱温27℃。结果:氨基酸浓度在2.50~125.0μg/mL范围内,线性相关系数均大于0.99;氨基酸的平均回收率在97%~106%之间。但山羊角提取物各批次之间氨基酸含量差异较大。结论:柱前衍生DNFB法适合用于山羊角提取物中氨基酸的检测。
Objective: To study the determination of amino acids in goat horn extract. Methods: Conventional amino acids in goat horn extract were detected by high performance liquid chromatography (HPLC) with 2,4-dinitrofluorobenzene (DNFB-HPLC). The mobile phase A: acetonitrile-water (1: 1), mobile phase B: 0.05 mol / L sodium acetate buffer (pH = 6.4) at a flow rate of 1.2 mL / min, detection wavelength 360 nm, column temperature 27 ℃. Results: The linear correlation coefficients were all over 0.99 in the range of 2.50 ~ 125.0 μg / mL. The average recoveries of amino acids ranged from 97% to 106%. However, the differences between the amino acid content of the goat horn extract batches larger. Conclusion: Pre-column derivatization DNFB method is suitable for the determination of amino acids in goat horn extract.
