以小桐子(Jatropha curcas)c DNA为模版,克隆了Jc NF-YB3和Jc NF-YB10基因的CDS序列。序列分析表明该两个基因分别包含549和528 bp完全阅读框(ORF),编码182和175个氨基酸。Blast搜索结果及进化分析结果表明,Jc NF-YB3和Jc NFYB10都与油桐(Vernicia fordii)NF-YB家族基因序列一致性最高且亲缘关系最近。定量分析表明,Jc NF-YB3和Jc NF-YB10在小桐子根、茎、叶、花、果皮和种子中都有表达,Jc NF-YB3在根中表达量最高,Jc NF-YB10在种子中表达量最高。两个基因在种子发育期的表达量变化趋势一致,但是Jc NF-YB10在种子整个生长发育期的表达量都要高于Jc NF-YB3。Jc NFYB3和Jc NF-YB10都在Na Cl、PEG、4°C低温和ABA处理后表达量不同程度上调,推测这两个基因在小桐子中参与抗逆反应机制和植物激素信号传导。
The CDS sequences of Jc NF-YB3 and Jc NF-YB10 genes were cloned using Jatropha curcas cDNA as a template. Sequence analysis showed that the two genes contained 549 and 528 bp complete ORFs respectively encoding 182 and 175 amino acids. Blast search results and phylogenetic analysis showed that both Jc NF-YB3 and Jc NFYB10 had the highest identity and closest relationship with the NF-YB family of Vernicia fordii gene sequences. Quantitative analysis showed that Jc NF-YB3 and Jc NF-YB10 were expressed in the roots, stems, leaves, flowers, pericarp and seeds of Jatropha curcas, Jc NF-YB3 was highest in roots, Jc NF-YB10 was in seeds The highest expression. However, the expression of Jc NF-YB10 was higher than that of Jc NF-YB3 during the entire growth and development of seeds. Both Jc NFYB3 and Jc NF-YB10 were upregulated to varying degrees after NaCl, PEG, 4 ° C hypothermia and ABA treatment, suggesting that these two genes are involved in anti-retrogradation mechanisms and plant hormonal signaling in Jatropha curcas.
