用ABC免疫组织化学染色法,观察猪卵透明带单克隆抗体(LPD8、LPC4),在体外与人、猪卵巢组织的反应。LPD8和LPC4使人、猪卵透明带及卵细胞质着色,但不使卵巢的其他结构,如颗粒细胞、卵泡膜细胞及结缔组织着色。这些结果提示:猪卵透明带单克隆抗体对卵巢内分泌功能可能无直接影响,但可过过与卵细胞质结合,抑制卵泡的发育,继而导致卵泡数目减少和内分泌功能降低。作者认为,用本实验提取的两株猪卵透明带单克隆抗体,进行免疫避孕研究前途不大。
ABC immunohistochemical staining was used to observe the reaction of porcine zona pellucida monoclonal antibody (LPD8, LPC4) with human and porcine ovarian tissue in vitro. LPD8 and LPC4 stain human zona pellucida and egg cytoplasm, but do not stain other structures of the ovary, such as granulosa cells, orca cells, and connective tissue. These results suggest that the porcine zona pellucida monoclonal antibody may not have a direct effect on the ovarian endocrine function, but may overexpress the cytoplasm and inhibit the development of the follicle, leading to the reduction of the number of follicles and the decrease of the endocrine function. The author believes that the use of the experimental two zona pellucida monoclonal antibodies, immunocontraceptive research has little prospect.
