[目的]研究脂质体介导质粒DNA转染293细胞的优化条件。[方法]通过改变DNA和脂质体的比例来进行优化,设为4个组,各组中DNA和脂质体的比例(μg∶μl)依次设定为1∶2.5、1∶3.5、1∶4.0和1∶5.0,以LacZ为报告基因,测定不同实验组的细胞转染率。[结果]当DNA/脂质体(μg∶μl)=1∶3.5时细胞转染率最高,1∶2.5(脂质体说明书推荐的比例)时细胞转染率最低,两者差异有统计学意义(P<0.05)。[结论]通过优化转染条件可以显著提高脂质体介导质粒DNA的转染率。
[Objective] The research aimed to study the optimal conditions of liposome-mediated plasmid DNA transfection into 293 cells. [Method] The DNA and liposome ratios (μg: μl) were optimized by changing the ratios of DNA and liposomes to 4 groups, and the ratio of DNA to liposome in each group was set to 1: 2.5, 1: 3.5, : 4.0 and 1: 5.0, with LacZ as the reporter gene to determine the cell transfection rate of different experimental groups. [Result] When the ratio of DNA / liposome (μg: μl) = 1: 3.5 was the highest transfection rate, the transfection rate was the lowest when the ratio of DNA / liposome was 1: Significance (P <0.05). [Conclusion] The transfection efficiency of liposome-mediated plasmid DNA can be significantly improved by optimizing transfection conditions.
