白藜芦醇致敏 TRAIL 诱导人肝癌细胞凋亡及其相关机制的研究

目的研究白藜芦醇对TRAIL诱导肝癌细胞SMMC-7721凋亡的影响及其机制。方法培养人肝癌细胞株SMMC-7721,分为对照组、TRAIL干预组、Res+TRAIL干预组,进行DNA末端原位标记(TUNEL)染色法作凋亡分析;Western Blot检测白藜芦醇对SMMC-7721细胞Survivn表达的影响。结果对照组SMMC-7721细胞的凋亡指数为(1.78±0.49)%,TRAIL干预组凋亡指数为(9.86±0.53)%,Res+TRAIL干预组中25μmol/L、50μmol/L、100μmol/L浓度Ros凋亡指数分别为(11.30±0.80)%,(20.89±0.69)%和(27.78±1.88)%。除Res+TRAIL干预组(25μmol/L)与TRAIL干预组比较无显著性差异(P>0.05),其余各组之间比较差异均有统计学意义(P均<0.05)。Western Blot测定结果显示:25μmol/L Ros处理组Survivin/β-actin的值与对照组相比无统计学差异(P>0.05);50μmol/L和100μmol/L Res处理组Survivin/β-actin的值较对照组显著降低(P<0.01);100μmol/L Res处理组Survivin/β-actin的值较50μmol/L显著降低(P<0.01)。结论 TRAIL对SMMC-7721细胞有凋亡诱导作用;Res对TRAIL诱导人肝癌细胞SMMC-7721凋亡有致敏作用,并且这种作用呈剂量依赖关系;其分子机制可能与下调凋亡抑制因子Survivin的表达有关。 Objective To study the effect and mechanism of resveratrol on apoptosis of hepatocellular carcinoma cell line SMMC-7721 induced by TRAIL. Methods Human hepatocellular carcinoma cell line SMMC-7721 was cultured and divided into control group, TRAIL intervention group and Res + TRAIL intervention group. TUNEL staining was used for apoptosis analysis. Western Blot was used to detect the effect of resveratrol on SMMC- -7721 cells Survivn expression. Results The apoptosis index of SMMC-7721 cells in control group was (1.78 ± 0.49)%, that in TRAIL intervention group was (9.86 ± 0.53)%, and that in Res + TRAIL group was 25μmol / L, 50μmol / L and 100μmol / L The apoptotic index of Ros was (11.30 ± 0.80)%, (20.89 ± 0.69)% and (27.78 ± 1.88)%, respectively. There was no significant difference (P> 0.05) except Res + TRAIL intervention group (25μmol / L) and TRAIL intervention group (P <0.05). The results of Western Blot showed that the Survivin / β-actin levels in 25μmol / L Ros treated group were not significantly different from those in control group (P> 0.05). Survivin / β-actin in 50μmol / L and 100μmol / (P <0.01). Survivin / β-actin in 100μmol / L Res group was significantly lower than that in 50μmol / L group (P <0.01). Conclusion TRAIL induces apoptosis in SMMC-7721 cells. Res induces TRAIL-induced apoptosis in SMMC-7721 cells in a dose-dependent manner. Its molecular mechanism may be related to the down-regulation of Survivin Expression related.
