目的 研究红毛五加多糖抑癌机制 .方法 采用DNA电泳检测红毛五加多糖对体外培养人胃癌细胞 DNA的影响 .结果 0 .1mm ol· L- 1 红毛五加多糖分别作用胃癌细胞 36 ,48和 6 0 h后 ,提取 DNA电泳呈现梯状图谱 ,且具有时间—效应关系 .至 72 h,电泳带型接近顺铂坏死对照组 ,只形成连续的条带 ,无 DNA梯状图谱 .Vit C对照组与空白对照组既无梯状图谱 ,也无连续条带 .结论 红毛五加多糖有诱导胃癌细胞凋亡的作用 .
OBJECTIVE To study the mechanism of polysaccharide inhibition by Acanthopanax senticosus polysaccharides. Methods DNA electrophoresis was used to detect the effect of Acanthopanax giraldii Harms polysaccharides on the DNA of human gastric cancer cells. The results showed that 0.1 mg ol·L-1 polysaccharides acted on gastric cancer cells respectively. After 48 hours and 60 hours, the DNA electrophoresis showed a ladder pattern with a time-effect relationship. At 72 h, the electrophoresis band shape was close to the cisplatin necrosis control group. Only a continuous band was formed and there was no DNA ladder pattern. Vit C control group and blank control group had neither laddering nor continuous bands. Conclusion Acanthopanax senticosus polysaccharide can induce gastric cancer cell apoptosis.
