目的:建立栀子中西红花苷-Ⅰ和西红花苷-Ⅱ的含量测定方法。方法:采用HPLC-DAD法,Dikma Diamonsil C18色谱柱(4.6 mm×250 mm,5μm),流动相0.1%甲酸水-0.1%甲酸乙腈梯度洗脱,流速1 mL.min-1,检测波长440 nm。结果:西红花苷-Ⅰ在10.272~256.8 mg.L-1线性关系良好;平均回收率96.68%,RSD 1.2%。西红花苷-Ⅱ在2.544~63.6 mg.L-1线性关系良好;平均回收率102.94%,RSD 0.87%。结论:该方法可靠,重复性好,可作为栀子质量控制的进一步补充。
Objective: To establish a method for the determination of crocin-I and crocin-II in gardenia. Methods: HPLC-DAD method was used on a Dikma Diamonsil C18 column (4.6 mm × 250 mm, 5 μm). The mobile phase was eluted with a gradient of 0.1% formic acid and 0.1% formic acid in acetonitrile with a flow rate of 1 mL · min- . Results: The linear relationship of caryol-I was 10.272 ~ 256.8 mg.L-1. The average recovery was 96.68% and the RSD was 1.2%. The linear relationship of crocin-II between 2.544 and 63.6 mg · L-1 was good with the average recovery of 102.94% and RSD 0.87%. Conclusion: The method is reliable, reproducible and can be used as a further supplement to the quality control of Gardenia.
