[目的]观察血管紧张素转化酶抑制剂(ACEI)依那普利对肾性高血压大鼠脑缺血再灌注后脑组织基质金属蛋白酶(MMP)-2、MMP-9表达的影响。[方法]肾性高血压大鼠模型采用两肾一夹法制备,大脑缺血再灌注模型采用大脑中动脉线栓法制备。将Wister雄性大鼠36只随机分为肾性高血压组和正常血压组,肾性高血压组再随机分为依那普利组(E组)和缺血再灌注组(HIR组),分别采用依那普利和0.9%氯化钠注射液灌胃4周。正常血压组再随机分为正常组(N组)和缺血再灌注组(IR组),IR组进行缺血再灌注处理。采用免疫组化法检测4组大鼠脑组织MMP-2、MMP-9的表达,用图像分析仪测定其灰度值。[结果]MMP-2、MMP-9灰度值IR组明显高于N组(P﹤0.01),HIR组高于IR组(P﹤0.05),E组明显低于HIR组(P﹤0.01)。[结论]高血压加重脑缺血再灌注后脑组织MMP-2、MMP-9的表达,ACEI能明显抑制脑组织MMP-2、MMP-9的表达。
[Objective] To observe the effect of enalapril, an angiotensin converting enzyme inhibitor (ACEI), on the expression of matrix metalloproteinase (MMP-2, MMP-9) in brain tissue in renovascular hypertensive rats after cerebral ischemia-reperfusion. [Method] Renal hypertensive rat model was prepared by two kidneys and one clip method. Cerebral ischemia-reperfusion model was prepared by middle cerebral artery occlusion method. Thirty-six male Wister rats were randomly divided into renal hypertension group and normal blood pressure group. Renal hypertension group was randomly divided into enalapril group (E group) and ischemia reperfusion group (HIR group) Enalapril and 0.9% sodium chloride injection for 4 weeks. The normal blood pressure group was randomly divided into normal group (N group) and ischemia reperfusion group (IR group). IR group was given ischemia reperfusion. Immunohistochemistry was used to detect the expression of MMP-2 and MMP-9 in brain tissue of four groups. The gray value was measured by image analyzer. [Result] The gray value of MMP-2 and MMP-9 in IR group was significantly higher than that in N group (P <0.01), HIR group was higher than IR group (P <0.05), E group was lower than HIR group . [Conclusion] Hypertension aggravates the expression of MMP-2 and MMP-9 in brain tissue after cerebral ischemia-reperfusion. ACEI can significantly inhibit the expression of MMP-2 and MMP-9 in brain tissue.
