目的:寻找新的TIL刺激物,摸索迅速解除TIL抑制状态和提高扩增效率的最佳条件。方法:通过黄芪、干地黄水提物联合细胞因子,采用3H-TdR掺入法及流式细胞仪对9例食管癌TIL的增殖及表型进行了分析。结果:在促增殖作用方面,黄芪联合IL-2、IL-2+IL-4、IL-2+TNF-a均较单用其细胞因子有明显增强作用。而干地黄对TIL增殖无明显协同作用。食管癌TIL表型分析发现CD4+T细胞为主,诱导后CD4+T细胞明显增多,CD4/CD8比值由2.8:1上升至9.9:1。结论:黄芪有明显协同细胞因子促进食管癌TIL增殖的作用,激活的食管癌TILCD4+T细胞占绝对优势。
OBJECTIVE: To search for new TIL stimulators and explore the best conditions for rapidly relieving TIL inhibition and increasing amplification efficiency. METHODS: The proliferation and phenotypes of TIL from 9 cases of esophageal cancer were analyzed by 3H-TdR incorporation method and flow cytometry using Astragalus membranaceus and Radix et Rhizoma water extract combined with cytokines. RESULTS: In the aspect of promoting proliferation, Astragalus combined with IL-2, IL-2 + IL-4, IL-2 + TNF-a were significantly enhanced by cytokines alone. However, dry yellow has no significant synergistic effect on the proliferation of TIL. The TIL phenotype analysis of esophageal cancer revealed that CD4+ T cells were predominant and CD4+ T cells increased significantly after induction. The ratio of CD4/CD8 increased from 2.8:1 to 9.9:1. Conclusion: Astragalus has a significant synergistic effect on cytokines to promote the proliferation of esophageal cancer TIL, and the activated esophageal cancer TILCD4+ T cells predominate.
