目的:测定不同采收时期牻牛儿苗中活性成分没食子酸、原儿茶酸、柯里拉京及鞣花酸的含量,为最佳采收期的确定提供依据。方法:采用RP-HPLC,AgilentTC-C18色谱柱(4.6 mm×250 mm,5μm),0.4%磷酸水溶液-甲醇为流动相梯度洗脱,流速0.8 mL·min-1,检测波长259 nm,柱温30℃。结果:没食子酸,原儿茶酸,柯里拉京,鞣花酸的线性范围分别为0.492~4.920 g·L-1(r=0.999 3),0.497~4.970 g·L-1(r=0.999 7),0.995~9.950 g·L-1(r=0.999 8),0.989~9.890 g·L-1(r=0.999 7);平均回收率分别为100.46%(RSD 2.1%),99.78%(RSD 1.3%),100.18%(RSD 1.0%),100.21%(RSD1.4%)。结论:牻牛儿苗中没食子酸及原儿茶酸的含量在五月末达到最大值,柯里拉京及鞣花酸的含量在六月末达到最大值。4种酚酸活性成分总量在六月末达到最高。
Objective: To determine the content of gallic acid, protocatechuic acid, corilagin and ellagic acid in the sesquiterpenne bucks of different harvesting times, and provide the basis for the determination of the best harvesting time. Methods: The mobile phase was eluted with 0.4% phosphoric acid aqueous solution and methanol on a AgilentTC-C18 column (4.6 mm × 250 mm, 5 μm) using RP-HPLC with a flow rate of 0.8 mL · min-1 and a detection wavelength of 259 nm. 30 ° C. Results: The linear ranges of gallic acid, protocatechuic acid, corilagin and ellagic acid were 0.492 ~ 4.920 g · L -1 (r = 0.999 3), 0.497 ~ 4.970 g · L -1 (r = 0.999 7 ), 0.995-9.950 g · L -1 (r = 0.999 8) and 0.989-9.890 g · L -1 (r = 0.999 7). CONCLUSION: The content of gallic acid and protocatechuic acid in Juglans mandshurica reached the maximum at the end of May, and the content of Coryllagin and ellagic acid reached the maximum at the end of June. The total amount of 4 phenolic acids reached the highest level at the end of June.
