点特异性的蛋白质光交联探针已经成为研究活细胞内蛋白质-蛋白质相互作用的关键工具。北京大学陈鹏课题组长期致力于发展基于遗传密码子拓展的蛋白质光交联技术。在之前的工作中他们设计了高效的光敏感非天然氨基酸交联探针(Di ZPK),能够广泛地用于捕捉活细胞内的蛋白-蛋白相互作用。最近,在上述工作的基础之上,该课题组发展了“可切割”的新一代光交联探针Di ZSe K。与原有探针相比,新一代的探针在高效捕捉蛋白相互作用的同时,其骨架中所含的硒原子能够在过氧化氢氧化条件下断裂,从而实现对光交联获得的“诱饵蛋白-靶标蛋白”复合物的分离。进一步的,他们发展了能够与靶标蛋白上产生的次硒酸基
Point-specific protein photo-cross-linking probes have become a key tool for studying protein-protein interactions in living cells. Peking University Chen Peng research group has long been committed to the development of genetic code based on the expansion of protein light crosslinking technology. In their previous work, they designed Di ZPK, a highly efficient light-sensitive non-natural amino acid probe that can be widely used to capture protein-protein interactions in living cells. Recently, on the basis of the above work, the group has developed Di ZSe K., a new generation of “cross-linkable” photocrosslinking probes. Compared with the original probe, a new generation of probe can efficiently capture the protein interaction while the selenium atoms contained in the skeleton can be broken under the hydrogen peroxide oxidation condition so as to realize the “ Bait protein - target protein ”separation of the complex. Further, they developed sub-seleno-acids that can be produced on the target protein
