目的:建立钙黄绿素脂质体包封率的测定方法。方法:采用阴离子交换树脂微型柱分离脂质体与钙黄绿素;紫外可见分光光度法测定钙黄绿素的含量,检测波长为484nm,计算包封率。结果:钙黄绿素浓度在1~10mg·L-1范围内线性关系良好(r=0.9999);阴离子交换树脂能够吸附浓度在0.50~5.00g·L-1之间的钙黄绿素,柱通过率为0%;脂质体能够通过阴离子交换树脂柱而不被吸附,柱通过率在94.5%以上。利用阴离子交换树脂柱可以使脂质体与钙黄绿素得到良好的分离。利用该方法测得的钙黄绿素脂质体包封率为29.3%,RSD为1.6%(n=6)。结论:阴离子交换树脂-微柱离心法可以作为钙黄绿素脂质体包封率的测定方法,该方法简便快速,结果重复性好。
Objective: To establish a method for the determination of entrapment efficiency of calcein liposomes. Methods: Liposomes and calcein were separated by anion exchange resin micro-column. The content of calcein was determined by ultraviolet-visible spectrophotometry. The detection wavelength was 484 nm. The entrapment efficiency was calculated. Results: The linearity of calcein in the range of 1 ~ 10 mg · L-1 was good (r = 0.9999). Anion exchange resin was able to adsorb calcein at the concentration of 0.50 ~ 5.00 g · L-1 and the column passing rate was 0 %; Liposomes can pass through the anion exchange resin column without being adsorbed, the column passing rate is above 94.5%. Liposomes can be well separated from calcein by anion exchange resin column. The calcein liposome encapsulation efficiency measured by this method was 29.3% with a RSD of 1.6% (n = 6). CONCLUSION: Anion exchange resin - micro column centrifugation can be used as the determination method for the entrapment efficiency of calcein liposomes. The method is simple and rapid with good repeatability.
