目的:建立高效液相色谱法测定酸水解后金银花药材中常春藤皂苷元和齐墩果酸的含量。方法:采用反相高效液相色谱法,色谱柱为 HiQ Sil C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相为乙腈-水(85:15),流速1 mL·min~(-1),柱温为室温,进样量10μL,检测波长210 nm。采用硫酸加热回流水解4 h,氯仿索氏回流提取皂苷元。结果:常春藤皂苷元和齐墩果酸的线性范围分别为1.15~28.90μg及1.02~25.50μg,平均回收率分别为98.9%(RSD=1.8%)及98.5%(RSD=2.1%)。结论:所建立的 HPLC 方法重复性好,专属性强,为金银花药材质量控制标准的建立提供科学依据。
Objective: To establish a HPLC method for the determination of ivy saponin and oleanolic acid in honeysuckle herbs after acid hydrolysis. Methods: The reversed-phase high performance liquid chromatography was used. The column was HiQ Sil C 18 column (250 mm×4.6 mm, 5 μm). The mobile phase was acetonitrile-water (85:15). The flow rate was 1 mL·min~. (-1), column temperature was room temperature, sample volume was 10 μL, and detection wavelength was 210 nm. Sulfuric acid was heated to reflux for 4 h, and Soxhlet was extracted by Soxhlet in chloroform. RESULTS: The linear ranges of ivy saponin and oleanolic acid were 1.15 to 28.90 μg and 1.02 to 25.50 μg, respectively. The average recoveries were 98.9% (RSD=1.8%) and 98.5% (RSD=2.1%), respectively. Conclusion: The established HPLC method has good repeatability and specificity, which provides scientific basis for the establishment of quality control standards for honeysuckle medicinal materials.
