本文报告了大肠杆菌不耐热肠毒素(ETEC-LT)基因探针检测霍乱弧菌肠毒素(Vc-ET)基因的分子杂交条件(Vc-ET杂交法),证实LT探针在实验条件下可以检测Vc-ET,最低检测菌量为4个活菌,比国内其它检菌法敏感十几倍至几千倍。探针的有效期为45~60天,待检标本经简单处理(DNA变性)后冰箱存放1~2个月仍不影响检测结果。对1986年某边境地区病人和环境中搜集分离到的50株O-Ⅰ群EI-Tor菌株作初步研究,证实全部为有毒性的0-Ⅰ群霍乱弧菌,未发现在自然条件下因基因突变而产生的无毒性O-Ⅰ群霍乱弧菌存在。
In this paper, we report the molecular hybridization (Vc-ET hybridization) method of detecting Escherichia coli heat-labile enterotoxin (ETEC-LT) gene probe to detect the Vc-ET gene. It was confirmed that under the experimental conditions Vc-ET can be detected, the minimum detection capacity of 4 live bacteria, sensitive than other domestic bacteria ten times to several thousand times. Probes valid for 45 to 60 days, to be seized specimens after simple processing (DNA denaturation) after the refrigerator storage for 1 to 2 months still does not affect the test results. A preliminary study on 50 strains of EI-Tor isolates collected from patients and environments in a certain border area in 1986 confirmed that they were all virulent strains of V. cholerae 0 - Non-toxic O-1 group produced by the mutation Vibrio cholerae exist.
