为了确定不同来源的平菇遗传差异,利用相关序列扩增多态性(sequence-related amplified polymorphism,SRAP)分子标记技术,对安徽地区的16个平菇品种的遗传多样性进行了分析。筛选出24对随机引物组合,获得了627条清晰条带,平均每对引物产生26.1条。多态性条带共189条,占总条带数的30.1%,平均每对引物组合得到7.88条多态性条带。依据SRAP标记聚类,在遗传相似系数0.65的水平上,供试的16个平菇品种分为2个大类:Ⅰ和Ⅱ。Ⅰ类分为2个亚类:A类和B类,Ⅱ类也分为2个亚类:C类和D类。其中A类和D类各包括4个品种,B类中有3个品种,C类中有5个品种,从而在DNA水平上证明了所研究的平菇种质资源的遗传多样性。
To determine the genetic diversity of Pleurotus ostreatus from different origins, the genetic diversity of 16 Pleurotus ostreatus cultivars in Anhui Province was analyzed by using sequence-related amplified polymorphism (SRAP) molecular markers. A total of 24 pairs of random primers were selected and 627 clear bands were obtained, with an average of 26.1 pairs per primer. A total of 189 polymorphic bands, accounting for 30.1% of the total number of bands, the average combination of each primer obtained 7.88 polymorphic bands. According to SRAP marker clustering, 16 Pleurotus ostreatus cultivars were divided into two groups Ⅰ and Ⅱ on the level of genetic similarity coefficient 0.65. Category I is divided into two subcategories: Category A and Category B, Category II is also divided into two subcategories: Category C and Category D. Among them, four species were included in each of category A and D, three in category B and five in category C, demonstrating the genetic diversity of the Pleurotus ostreatus germplasm resources studied at the DNA level.
