目的:研究丹参对TGFβ1刺激的NIH/3T3成纤维细胞功能的影响。方法:将正常大鼠进行丹参灌胃,分离含药血清,温育TGFβ1刺激的NIH/3T3成纤维细胞.RT-PCR法检测c-fos基因表达,Gel mobility shift assay法检叙API蛋白的结合活性。结果:经TGFβ1刺激后,细胞c-fos mRNA 表达及 AP1蛋白结合活性明显增强,丹参含药血清可分别抑制由TGFβ1引起的细胞c-fos mRNA表达及API蛋白结合活性的增强。结论:丹参可以抑制TGFβ1刺激的NIH/3T3成纤维细胞中的c-fos基因表达及API蛋白结合活性。
Objective: To study the effects of salvia miltiorrhiza on TGFβ1-stimulated NIH/3T3 fibroblasts. METHODS: Normal rats were intragastrically administered Salvia miltiorrhiza, serum containing drug was isolated, and TGFβ1-stimulated NIH/3T3 fibroblasts were incubated. The expression of c-fos gene was detected by RT-PCR. Gel mobility shift assay was used to detect the binding of API protein. active. RESULTS: After stimulated by TGFβ1, the expression of c-fos mRNA and the binding activity of AP1 protein were significantly enhanced. Salvia miltiorrhiza drug serum inhibited the expression of c-fos mRNA induced by TGFβ1 and enhanced the protein binding activity of API. Conclusion: Salvia miltiorrhiza can inhibit c-fos gene expression and API protein binding activity in TGFβ1-stimulated NIH/3T3 fibroblasts.
