目的探讨α-硫辛酸(α-LA)对糖尿病大鼠肾脏聚ADP-核糖多聚酶(PARP)表达的影响。方法 30只SD大鼠随机均分为链脲佐菌素(STZ)诱导的糖尿病组(A组)、α-LA干预的糖尿病组(B组)及正常对照组(C组)。8周后测各组大鼠肾脏超氧化物歧化酶(SOD)、丙二醛(MDA)含量,TUNEL法检测肾脏细胞凋亡,免疫组化法观察PARP在肾脏中的表达,Western blot法检测肾脏半胱氨酸蛋白酶3(Caspase-3)的表达。结果与C组比,A组大鼠肾脏SOD活性下降,MDA含量、肾脏细胞凋亡数、PARP和Caspase-3的表达均显著增多(P<0.05),而B组上述指标均较A组明显改善(P<0.05)。结论α-LA对糖尿病大鼠肾脏保护作用可能与抑制氧化应激、下调PARP表达而对抗细胞凋亡有关。
Objective To investigate the effect of α-lipoic acid (α-LA) on the expression of poly ADP-ribose polymerase (PARP) in the kidney of diabetic rats. Methods Thirty SD rats were randomly divided into three groups: diabetic group (group A) induced by streptozotocin (STZ), diabetic group (group B) treated with α-LA, and normal control group (group C). After 8 weeks, the contents of superoxide dismutase (SOD) and malondialdehyde (MDA) in kidney of rats in each group were measured. The apoptosis of renal cells was detected by TUNEL method. The expression of PARP in kidney was observed by immunohistochemical method. Kidney cysteine proteinase 3 (Caspase-3) expression. Results Compared with group C, the activity of SOD, the content of MDA, the number of apoptotic cells and the expression of PARP and Caspase-3 in group A were significantly increased (P <0.05), while those in group B were significantly higher than those in group A Improve (P <0.05). Conclusion The protective effects of α-LA on the kidney of diabetic rats may be related to inhibition of oxidative stress, down-regulation of PARP and anti-apoptosis.
