目的:从外周淋巴细胞比例、基因和血清细胞因子的角度研究补肾通督胶囊对胶原诱导关节炎大鼠TH1/TH2细胞的影响。方法:将42只大鼠,分为正常对照组(C)、模型组(M)、补肾通督胶囊的低剂量组(BSL)、中剂量组(BSM)、高剂量组(BSH)和雷公藤组(TG)。第1天进行造模,从第13天开始灌胃18d,在第31天取材,进行流式细胞术、RT-PCR和ELISA检测Th1和Th2细胞、IL-4和IFN-γ的基因表达及血清水平。结果:补肾通督组的Th1的百分比随着剂量的提高有下降的趋势,而Th2的百分则呈现上升的趋势。随着补肾通督剂量的提高,IFN-γ的基因表达和血清水平呈现下降的趋势,而IL-4的基因表达和血清水平则有上升的趋势。结论:补肾通督随着剂量的提高,不但降低外周血中Th1细胞的表达,同时提高Th2细胞的表达,对IL-4和IFN-γ的基因表达和血清水平皆有抑制Th1细胞效应和提高Th2细胞效应的趋势,提示其缓解RA的机制与调控Th细胞的分化与平衡密切相关。
OBJECTIVE: To study the effects of Bushen Tongdu capsule on TH1 / TH2 cells in collagen-induced arthritis rats from the perspective of peripheral lymphocyte ratio, gene and serum cytokines. Methods: Forty-two rats were divided into normal control group (C), model group (M), low dose group (BSL), medium dose group (BSM), high dose group (BSH) Rattan group (TG). On the first day, the model was established. On the 13th day, the mice were gavaged for 18 days. On the 31st day, the cells were harvested for flow cytometry. The gene expressions of Th1 and Th2 cells, IL-4 and IFN-γ were detected by RT-PCR and ELISA. Serum levels. Results: The percentage of Th1 in Bushen Tongdu group tended to decrease with the increase of dose, while the percentage of Th2 tended to increase. With the increase of Bushen Tongxue dose, the gene expression and serum level of IFN-γ showed a downward trend, while the gene expression and serum level of IL-4 showed an upward trend. Conclusion: With the increase of dose, Bushen Tongsuo can not only reduce the expression of Th1 cells in peripheral blood, but also increase the expression of Th2 cells. Both the gene expression and serum levels of IL-4 and IFN-γ inhibit the effect and increase of Th1 cells Th2 cell trend, suggesting that its mechanism of RA and regulation of Th cell differentiation and balance are closely related.
