目的:观察匙羹藤(Gymnema sylvestre,GS)对自发性2型糖尿db/db小鼠空腹血糖(FBG)、血脂(TG、TC、LDL、HDL)、血清胰岛素(Fins)及脂联素(ADPN)、TNF-αmRNA表达的影响,探讨GS对胰岛素抵抗的作用及对ADPN、TNF-α基因表达的关系。方法:将SPF级27只db/db小鼠随机分3组(n=9):模型组、GS组、吡格列酮组;并选8只db/+m鼠为正常组。连续灌胃给药4周后检测FBG、Fins、血脂、FFA,并用RT-PCR检测脂肪组织中ADPN、TNF-αmRNA的表达。结果:模型组和治疗组的体质量、FBG、Fins、TG、CHOL、LDL、FFA水平和TNF-α的mRNA表达明显高于正常组,而ISI指数和ADPN mRNA的表达低于正常组;与模型组相比,匙羹藤组db/db小鼠的FBG、Fins、TC、LDL-C水平降低,提高ISI指数,上调ADPN mRNA的表达。结论:匙羹藤可改善糖脂代谢紊乱,增加胰岛素的敏感性,其机制可能与改善脂肪细胞因子TNF-α和ADPN的表达有关。
Objective: To observe the effects of Gymnema sylvestre (GS) on fasting blood glucose (FBG), blood lipids (TG, TC, LDL, HDL), serum insulin and adiponectin ADPN) and TNF-αmRNA, to explore the effect of GS on insulin resistance and its relationship with the expression of ADPN and TNF-α. Methods: 27 db / db mice of SPF grade were randomly divided into 3 groups (n = 9): model group, GS group and pioglitazone group. Eight db / + m mice were selected as normal group. FBG, Fins, lipids and FFA were detected after continuous gavage for 4 weeks. The expression of ADPN and TNF-αmRNA in adipose tissue was detected by RT-PCR. Results: The body mass, FBG, Fins, TG, CHOL, LDL, FFA and the mRNA expression of TNF-α in model group and treatment group were significantly higher than those in normal group, while those in ISI index and ADPN mRNA were lower than those in normal group Compared with model group, FBG, Fins, TC, LDL-C levels of Gymnema sylvestre-treated db / db mice decreased, ISI index increased and ADPN mRNA expression increased. Conclusion: Gymnema sylvestre can improve glucose and lipid metabolism disorders and increase insulin sensitivity, the mechanism may be related to the improvement of the expression of adipocyte cytokines TNF-α and ADPN.
