目的:探讨人参皂苷Rb1(GS Rb1)促进小鼠骨髓基质细胞(BMSCs)生长及干预长春新碱(VCR)抑制小鼠BMSCs增殖的作用。方法:正常对照组,GS Rb1(1.0、5.0、10.0、15.0μg/ml)诱导BMSCs增殖组,VCR(2.5、5.0、10.0、15.0μg/ml)抑制BM-SCs增殖组,GS Rb1干预VCR抑制BMSCs增殖组,应用MTT法测定BMSCs增殖。结果:①与正常对照组比较,GS Rb1 1.0~10.0μg/ml随浓度升高其促进BMSCs增殖也增加,GS Rb1为10.0μg/ml时差异具有显著性(P<0.05);VCR 2.5~15.0μg/ml随浓度的升高抑制BMSCs增殖也增强,VCR 5.0~15.0μg/ml时差异具有显著性(P<0.05);GS Rb1 1.0~15μg/ml各组与VCR 5.0~15.0μg/ml各组比较,以及GS Rb1 10.0μg/ml组与VCR 2.5μg/ml组比较其BMSCs增殖显著增加(P均<0.05)。②GS Rb1 5.0μg/ml、10.0μg/ml1、5.0μg/ml分别与VCR 5.0μg/ml培养,其能显著干预VCR对BMSCs增殖的抑制(P均<0.05)。结论:长春新碱明显抑制骨髓基质细胞生长;人参皂苷Rb1可促进骨髓基质细胞生长且具有干预长春新碱对骨髓基质细胞增殖的抑制作用。
Objective: To investigate the effects of ginsenoside Rb1 (GS Rb1) on the growth of mouse bone marrow stromal cells (BMSCs) and the effect of vincristine (VCR) on the proliferation of mouse BMSCs. Methods: The proliferation of BMSCs was induced by GS Rb1 (2.5, 5.0, 10.0 and 15.0 μg / ml) in the normal control group. The proliferation of BM-SCs was inhibited by VCR BMSCs proliferation group, MTT assay BMSCs proliferation. Results: ① Compared with the normal control group, GS Rb1 1.0 ~ 10.0μg / ml increased the proliferation of BMSCs with increasing concentration of GSRb1, the difference was significant (P <0.05) when GS Rb1 was 10.0μg / ml; VCR 2.5 ~ 15.0 μg / ml inhibited the proliferation of BMSCs with the increase of concentration, the difference was significant at VCR 5.0 ~ 15.0μg / ml (P <0.05); GS Rb1 1.0 ~ 15μg / ml and VCR 5.0 ~ 15.0μg / ml Compared with the control group, the proliferation of BMSCs in GS Rb1 10.0μg / ml group and VCR 2.5μg / ml group was significantly increased (all P <0.05). ② GS Rb1 5.0μg / ml, 10.0μg / ml1,5.0μg / ml cultured with VCR 5.0μg / ml, respectively, could significantly inhibit the proliferation of BMSCs inhibited by VCR (all P <0.05). CONCLUSION: Vincristine significantly inhibits the growth of bone marrow stromal cells. Ginsenoside Rb1 can promote the growth of bone marrow stromal cells and inhibit the proliferation of bone marrow stromal cells by vincristine.
